Why MUST it be 40, 45, or 10mm?
In general, I'd suggest a S&W M&P 40 w/ good night sights... for $500... not $5000! I assume you mean $500.
But there's LOTS of options.

Are you planning on attending a gun fight?
Pick a caliber that can get the job done and a pistol that you can shoot accurately which has earned your confidence. We aren't going to get into a brand/model comparison here. There is plenty of information on the boards about which pistol(s) are most likely to be found in the holsters of those who take such things seriously here.
